Relative quantitative information about components of chemical or biological samples can be obtained from mass spectra by normalizing the spectra to yield peak intensity values that accurately reflect concentrations of the responsible species. A normalization factor is computed from peak intensities of those inherent components whose concentration remains constant across a series of samples. Relative concentrations of a component occurring in different samples can be estimated from the normalized peak intensities. Unlike conventional methods, internal standards or additional reagents are not required. The methods are particularly useful for differential phenotyping in proteomics and metabolomics research, in which molecules varying in concentration across samples are identified. These identified species may serve as biological markers for disease or response to therapy.

A method of producing a homogenous population of homozygous stem (HS) cells pre-selected for immunotype and/or genotype from donor cells is described herein. The invention relates to methods of using immunohistocompatible HS cells for diagnosis, therapeutic and cosmetic transplantation, and the treatment of various genetic diseases, neurodegenerative diseases, traumatic injuries and cancer. The invention further relates to methods for using histocompatible HS stem cells pre-selected for a non-disease genotype for prophylactic and therapeutic intervention including, but not limited to, therapeutic and cosmetic transplantation, and the treatment of various genetic diseases, neurodegenerative diseases, and cancer. Furthermore, the invention relates to a catalogued transplant depository of HS cells derived from multiple donors, each of the HS cells being homozygous for a unique HLA haplotype, for the purpose of having a constant, reliable, comprehensive supply of immunohistocompatible cells for diagnosis, treatment and/or transplantation.

The present invention relates to materials and methods for the quantitative and qualitative analysis of gene expression during targeted differentiation of isolated homozygous stem cells, and to materials and methods for accumulating sequence tags sampled from a population of expressed genes. More particularly, the present invention relates to a method for constructing a model of cellular development and differentiation comprising the steps of: (1) producing a homogenous population of homozygous stem (HS) cells; (2) directing the differentiation of the HS cells to arrive at desired mature (somatic) cells, group of cells or tissue; (3) periodically sampling HS cells undergoing differentiation to extract total cellular RNA produced at various sampling stages; (4) isolating mRNA from the total cellular RNA; and (5) constructing a cDNA library representative of proteins produced and expressed at various stages of differentiation, over the course of time. The present invention further relates to sequencing and cataloguing isolated mRNAs and cDNAs at various stages of differentiation of HS cells, producing and cataloguing isolated polypeptides sequences encoded by same, and screening the cDNA libraries. In vitro and in vivo cultures of homozygous stem cell are also described; such cultures find utility both as models of differentiation as well as sources of differentiated tissues and organs.
