Methods are described for attaching molecules to a solid phase, such as a magnetic bead. Free amine groups on the solid surface react with a molecule that contains a sulfhydryl group. The sulfhydryl group may, but need not be protected. The molecule is then reacted with a conjugate of linker and ligand. The liner has a reactive maleimide or sulfhydryl group which reacts with the molecule on the solid phase.

Abstract:
Disclosed herein are architectures for an external cavity laser. In some embodiments, the external cavity laser includes vertical cavity surface emitting laser (VCSEL) elements, a Brewster plate, frequency doubling chips, and a microlens array. The Brewster plate is arranged at an angle relative to the light path, and is configured to polarize at least the light received from the VCSELs and propagating on the light path in a first direction, and extract, from the external cavity, frequency-doubled light propagating on the light path in a second direction opposite to the first direction. The doubling chips are operable to receive the light and double the frequency of a portion of the received light. The microlens array is aligned with the VCSEL elements. A mount may be employed to mount the side stack of doubling chips by either side mounting or end mounting.
A method for characterizing a target peptide through a detection approach such as mass spectrometry is provided, including: introducing at least one guard molecule to mix with the target peptide; and applying the detection approach for the characterization of the target peptide. Each guard molecule is configured to have similar characteristics as the target peptide, yet is still distinguishable therefrom by the detection approach, such as having a mass spectrometry-distinguishable different M/z value compared with the target peptide. The method can be used to characterize a neoantigen peptide through mass spectrometry, upstream of which the method can further include steps for tissue sample preparation, HLA molecules enrichment, elution, clean-up, and purification. Some or all of these steps can be configured to be executed in a substantially automatic manner with little or no manual intervention. A system for implementing the neoantigen analysis method is further provided.

Abstract:
A computer implemented method for identifying an application topology includes identifying a sandbox environment corresponding to an application of interest, analyzing the sandbox environment to identify a set of communication links between services within the sandbox environment indicating a first topology, identifying a production system corresponding to the application of interest, querying the production system to identify a set of structural dependencies indicating a second topology, and creating a complete topology of the cloud application by combining the first topology and the second topology. A computer program product and computer system for identifying an application topology are additionally disclosed herein.
Error-Proof Nucleic Acid Library Construction Method
A method and kit for constructing a barcoded single-stranded DNA library are disclosed. The method includes preparing single-stranded DNA molecules each having a dephosphorylated 5′ end, ligating a first adaptor to a 3′ end of each single-stranded DNA molecule, and synthesizing a complementary strand of each single-stranded DNA molecule ligated to the first strand of the first adaptor. The kit includes the first adaptor having a first strand, which includes, from a 5′ end to a 3′ end, a phosphate group, a barcode sequence, and a first primer recognition sequence. The kit also includes a DNA ligase for a ligation between the 5′ end of the first strand of the first adaptor to each single-stranded DNA molecule, and a first primer for the synthesis of the complementary strand. The method allows for analysis of rare mutations and from nucleic acid samples of low quality and quantity.
A method and a kit for enriching target nucleic acid sequences from a biological sample are disclosed. The method includes preparing, and contacting with the biological sample, a first RNA probe set and a second RNA probe set respectively and concurrently targeting both of the two antiparallel strands of a duplex segment in each target nucleic acid sequence. Each RNA probe in the first and second RNA probe set can be generated by chemical synthesis or by in vitro or in vivo transcription, and can be biotin-labelled to thereby allow capturing of the target nucleic acid sequences by magnetic beads labelled with streptavidin, or can be engineered to a microfluidic channel to facilitate the capturing. The method can be applied to capture double-stranded nucleic acid sequences or single-stranded nucleic acid sequences having duplex segments, and the nucleic acid sequences can include DNAs, RNAs, or DNA-RNA hybrid molecules.

A computer system, non-transitory computer storage medium, and a computer-implemented method of assessing technical risk using visual pattern recognition in an Information Technology (IT) Service Management System. A data visualization engine and a time series generation engine receive the operational data, respectively. A first representation of the data is generated by the data visualization engine, and a second representation of the data is generated by the time series generation engine. Anomaly patterns are identified by a pattern recognition engine configured to perform feature extraction and data transformation. An ensembler is configured to accept the outputs from two AI anomaly engines and make a final decision of whether anomaly patterns are captured. Risk scores based on the identified anomaly patterns are output by a pattern recognition engine to an automated management system. The anomalies includes information regarding vulnerabilities of devices or components of the IT Service Management System.
