Abstract:

The present disclosure provides systems and methods for making a hydrogel comprising a cell, cell nucleus, or one or more components derived from a cell or cell nucleus. A method for making a hydrogel may comprise providing a cell or cell nucleus, a first polymer, wherein the first polymer comprises a plurality of first crosslink precursors, each of the plurality of first crosslink precursors comprising an azide group; providing a second polymer, wherein the second polymer comprises a plurality of second crosslink precursors, each of the plurality of second crosslink precursors comprising an alkyne group; and crosslinking the first polymer and the second polymer via a reaction between a first section of the first crosslink precursors and a second section of the second crosslink precursors, thereby providing the hydrogel comprising the cell or cell nucleus.

Abstract:

A calibration device is disclosed. The device comprises an array of teaching pendants. The device comprises a translation actuator configured to translate the array of teaching pendants to a set of x and y positions, wherein the x and y positions are measured in a plane substantially parallel to a floor of an instrument deck. The device comprises a plurality of height actuators configured to move each of the teaching pendants in a direction substantially perpendicular to the plane. One or more of the teaching pendants contact one or more teaching objects of an array of teaching objects above the instrument deck as a result of a position of the array of teaching pendants.

Abstract:

Provided in some aspects are methods for light-controlled in situ surface patterning of a substrate. Compositions such as nucleic acid arrays produced by the methods are also disclosed. In some embodiments, provided herein is photocontrollable hybridization, where oligonucleotides with photo-caged bases prevent hybridization of complementary nucleic acid strands. Uncaging of the oligonucleotides allows hybridization and/or ligation of nucleic acid molecules at the exposed area. A large diversity of barcodes can be created in molecules on the substrate via sequential rounds of light exposure, hybridization, and ligation.

The present disclosure provides compositions, methods, systems, and devices for polynucleotide processing and analyte characterization. Such polynucleotide processing may be useful for a variety of applications, including analyte characterization by polynucleotide sequencing.
