Proteolytic enzyme compositions and processes for digesting connective tissue are disclosed. The enzyme compositions include collagenase, which is essentially free of toxins and non-collagen specific components, and chymopapain, which is essentially free of toxins. The enzyme compositions are used for dissociating microvessel cells from connective tissue. Recovered microvessel cells are incorporated into artificial vessel grafts. The enzyme compositions preferably contain an aqueous mixture of collagenase having an activity of about 43 nkat/ml to about 51 nkat/ml, and chymopapain having an activity of about 0. 22 nkat/ml to about 0. 44 nkat/ml.

Abstract:
A process for purifying crude collagenase is disclosed. The collagenase purification process includes providing a stabilized crude collagenase solution containing collagenase, pigment, toxins, bacterial materials, and proteolytic enzyme impurities including clostripain, trypsin, and caseinase. The stabilized collagenase solution is applied to hydroxylapatite packing. pigment and caseinase are eluted with a first solution comprising about 0. 05 M to about 0. 3 M phosphate buffer, and then collagenase, trypsin, and clostripain are eluted with a second solution comprising about 0. 35 M to about 0. 5 M phosphate buffer to provide a first collected solution. The first collected solution is then applied to gel filtration packing and collagenase and clostripain are eluted with a neutral pH buffer solution, to provide a second collected solution. The second collected solution is then applied to Reactive Red 120-Agarose packing and collagenase is eluted with a neutral pH buffer solution to provide purified collagenase. The process provides extremely pure collagenase in high yield with reduced consumption of eluting solutions and avoids unpredictable gradient eluting techniques.

The activities of chymopapain or papain in enzyme mixtures are inhibited by the addition of glycogen, hyaluronic acid, or desulfated heparin. The process for isolating viable hepatocytes or pancreatic cells by treatment with papain or chymopapain is improved by terminating the treatment by inhibiting the reaction with these polysaccharides. Methods for binding heparin to biological or synthetic materials which are to be implanted within a mammalian body. In instances where connective tissue proteins or other components of the material having adequate carboxyl groups present thereon, the method comprises a) contacting the material with a carboxyl-activating agent, b) contacting the material with a polyamine compound to form amide-bound polyarnine side chains at the sites of the previously activated carboxyl groups, and c) contacting the material with heparin such that heparin will become bound to the amino groups on the polyamine side chains. In other applications wherein the connective tissue proteins or other molecular entities of the material are devoid of or deficient in carboxyl groups, the method will comprise a) contacting the material with a carboxyl-activating agent in combination with a polyamine compound which has functional carboxyl groups thereon such that at least some of the functional carboxyl groups of the polyamine compound will become activated and will react with some of the amino groups on the polyamine compounds to form a polyamine network interlocked within the molecular structure of the material, and b) contacting the material with heparin such that heparin will become bound to amino groups on the polyamine network.
