Abstract:
A method of identifying and quantifying small RNA molecules comprising a) isolating RNA molecules; b) ligating RNA adapter molecules onto the isolated RNA molecules to form RNA template molecules; c) forming complementary DNA molecules by transcribing the RNA template molecules; d) amplifying the complementary DNA molecules; e) obtaining sequence information of the complementary DNA molecules (and thereby the RNA from which it was derived); and f) obtaining quantity information of the complementary DNA molecules, wherein the quantity information of the DNA molecules reflects the quantity of the isolated RNA molecules is provided. Included in the invention is the identification of RNA molecules between 15 and 30 nucleotides in length.

Genome edited plants, plant cells, seeds and plant parts of are provided where expression levels and/or activities of pod dehiscence genes are modulated to improve one or more agronomic characteristics such as pod shatter. Also provided are compositions comprising polynucleotides encoding polypeptides and guide RNAs targeted to endogenous proteins involved in pod dehiscence including for example, targeted site-directed mutagenesis using CRISPR-associated nucleases. Additionally, various methods of employing the polynucleotides and genetic modifications in plants, such as methods for modulating expression level in a plant and methods for increasing pod shatter tolerance of a plant are also provided herein.

Abstract:
Soybean seeds with increased protein and having a modified expression or activity of at least one or two HECT E3 ligase polypeptides are provided. Methods for modifying expression or activity of HECT E3 ligase polypeptides and polynucleotides include genome editing to modify the transcription regulatory region or sequence encoding the HECT E3 ligase polypeptides and transformation with recombinant DNA constructs to enhance or suppress expression or activity of the HECT E3 ligase polypeptides. Plants containing the modifications produce seeds with altered composition such as one or more of increased protein, decreased soluble carbohydrate, increased oleic acid, decreased saturated fats such as palmitic and stearic acids, and decreased linoleic or linolenic acid.
