Methods and kits are provided for the simultaneous detection of multiple analytes, i. e. to the determination of whether one or more of a plurality of analytes is present in a sample, particularly the plurality of analytes are structurally unrelated or significantly different, particularly to whether one or more of such analytes is present in a sample in a concentration that is above a predetermined minimum or cutoff value, and particularly for such analytes with different cutoffs. The methods and kits are particularly useful for-screening for the presence of a plurality of drugs (licit and/or illicit), performance-enhancing substances, and other chemicals, and involve a competitive enzyme immunoassay employing one or more conjugates of G6PDH with a plurality of the analytes.
Immunoassay For Specific Determination Of S-Adenosylmethionine And Analogs Thereof In Biological Samples
This invention pertains to a method for detecting a compound in the presence of other compounds that are substantially similar in structure and metabolically related to the analyte. The invention is particularly suited for the detection of S-adenosylmethionine in the presence of S-adenosylhomocysteine, other nucleosides and derivatives in a biological sample. The methods of this invention involve an antibody produced specifically against S-adenosylmethionine; particularly, analogs modified strategically at the sulfonium position. An assay protocol comprises chemically modified analyte analog linked to an enzymatic reporter and the aforementioned antibody was used to demonstrate the assay specificity and sensitivity. Additional assay method with immobilized immunogen, the specific antibody, and an enzyme labeled secondary antibody was also described for illustration. The invention also features hapten design and novel compounds used as haptens to prepare immunogen and for the specific antibody production.

Abstract:
A method is provided for determining the presence in a sample of a member of a specific binding pair ("sbp member") consisting of ligand and its homologous receptor. The sample is combined in an aqueous medium with (1) a complementary sbp member wherein at least the sbp member or the complementary sbp member is bound to the surface of a cell and (2) a fuorescent agent capable of being incorporated into the cell. The presence of the sbp member is indicated by a change in fluorescence of the unseparated cell suspension as a result of agglutination of the cells. The present invention has particular application to blood typing, for example, for the determination of the presence of blood group antigens A, B, AB, O, and D (Rh. sub. o) and antibodies to such antigens.
