Abstract:

A method for quantifying nucleic acid is provided. The method includes determining a first reference threshold cycle for a first predetermined input quantity for a reference nucleic acid, determining a first target threshold cycle for the first predetermined input quantity for a target nucleic acid, determining a second reference threshold cycle for a second predetermined input quantity for the reference nucleic acid, and determining a second target threshold cycle, by the processor, for the second predetermined input quantity for the target nucleic acid. The method further includes receiving a sample threshold cycle, determining a sample input quantity based on the first and second reference threshold cycle and the first and second target threshold cycle, and displaying the sample input quantity to a user.

Abstract:

A method of detection of a target nucleic acid is provided. The method includes fractionating a sample into a plurality of sample volumes wherein more than 50% of the fractions contain no more than 1 target nucleic acid molecule per sample volumes, and subjecting the plurality of sample volumes to conditions for amplification. The method further includes detecting a change in ion concentration in a sample volume wherein a target nucleic acid is present, counting the number of fractions with an amplified target nucleic acid, and determining the quantity of target nucleic acid in the sample.

Abstract:

This specification generally relates to methods of detecting the linkage between two or more targets in a sample using digital multiplex PCR. A method of identifying physical linkage between two or more nucleic acid targets in a sample is provided. The method includes diluting the sample via limiting dilution and aliquoting the diluted sample into wells. The method further includes performing multiplex PCR in each chamber, where a first dye is used for the first target and a second dye is used for the second target. The method includes identifying the presence of the first and second dyes in the wells, wherein a non-random distribution of the dyes identifies the targets as linked.
