Abstract:

The present invention provides a method of detecting a nuclease-mediated cleavage of a target nucleic acid through hybridizing a target nucleic acid to a fluorescently labeled oligonucleotide probe complementary to the target nucleic acid and containing a flourophor at one terminus and a quenching group at the other terminus. When the probe is unhybridized to the target nucleic acid, the probe adopts a conformation that places the flourophor and quencher in such proximity that the quencher quenches the flourescent signal of the flourophor and formation of the probe-target hybrid causes sufficient separation of the flourophor and quencher to reduce quenching of the flourescent signal of the flourophor. Once hybrized, the method contacts the probe-target hybrid with an agent having nuclease activity in an amount sufficient to selectively cleave the target nucleic acid and thereby release the intact probe. Detecting the release of the probe is then measured by following a decrease in the flourescent signal of the flourophor as compared to the signal of the probe-target hybrid.

Abstract:

A lace or draw string fastening system can include a clamping unit and a capture element. The clamping unit can clamp onto the laces, such as shoe laces, to tighten the shoes and the capture element can capture and contain the free ends of the laces that extend from the clamping unit to prevent tripping by securing them to other portions of the laces or the article that houses the laces. The fastening system can be used in a shoe lace fastening system without the need for knot tying. The clamping unit can clamp the laces together at the top of the shoe to hold the laces tight while the capture element can contain or secure the free ends of the shoe lace to the shoe or the laces adjacent the toe to prevent tripping.

Abstract:

A longitudinal cutting tool is formed with at least one groove and a stop collar is received on the cutting tool. The stop collar has a stop surface, and yieldably resilient means in the form of a threaded screw passing through a slot in the collar, and rubberized buttons inserted into the collar are used to adjust the collar on the cutting tool, and to operatively connect it thereto so as not to damage its groove; the cutting depth of the tool is limited when the stop face engages with the stop surface.

An electronic postage meter for printing postage indicia having a postage value and a message, in which the message is stored in a memory and the printer of the postage meter prints the message in accordance with that stored in the memory.
