Abstract:
A method and apparatus for red blood collection and filtration is provided wherein a red blood cell collection assembly provides for leukoreduction filtration concurrent with or soon after the red blood cell collection procedure. The procedure preferably involves filtering the separated red blood cells in a high hematocrit (high-crit) state prior to addition of storage solution thereto. Preferably, a storage solution is passed through the leukoreduction filter after the RBCs have been filtered therethrough. The red blood cell collection filtration and storage assembly is preferably preconnected to a blood component separation disposable assembly, including, for example, a centrifuge vessel and a blood removal/return assembly for removing blood from a donor, passing the blood to the centrifuge vessel for separation of the blood into components for collection and providing for filtration of the separated red blood cell component, as described.

Abstract:
Methods, devices and device components are presented for uniformly treating fluids undergoing mixing with electromagnetic radiation. In one aspect, the present invention provides methods of treating a fluid undergoing continuous fluid mixing wherein net radiant energies necessary to provide uniform treatment of the fluid samples with electromagnetic radiation are calculate on the basis of the volume, mass or mixing rate of the fluid or any combination of these variables. In another aspect, the present invention provides algorithms for determining net radiation energies, radiant powers, and/or illumination times necessary for providing uniform treatment of fluid samples. The present invention provides methods for uniformly reducing pathogens in biological fluids, including blood and blood components.
Antimicrobial Blood Treatment Using Allicin And Related Compounds
Methods and compositions prolonging the storage life and/or increasing the safety of a blood product, such as whole blood, red blood cells, white blood cells, platelets, serum and aqueous additive solutions for storage of such blood products are provided. Storage solutions of this invention comprise a composition selected from the group consisting of garlic extract, allicin, other microorganism-growth-inhibiting compounds derived from garlic, and analogs and derivatives of allicin and said other compounds, in an amount effective to inhibit growth of at least one selected microorganism which is a bacterium, virus, fungus or parasite. The storage additive solutions of this invention can increase platelet storage life by at least about 20%, preferably at least about 40%.
Methods for decontaminating fluids such as blood products, particularly platelets or red blood cells, are provided. The methods include mixing substantially non-toxic amounts of an endogenous photosensitizer such as riboflavin, or an endogenously-based derivative photosensitizer, with the fluid, increasing the dissolved oxygen content of the fluid to an amount sufficient to enhance a reaction of the photosensitizer in which singlet oxygen is formed and reduce competing reactions; and exposing the fluid to visible light photoradiation to activate the photosensitizer and substantially inactivate the pathogens. Blood products produced by the method, and systems for carrying out the method are also provided.

Abstract:
This invention relates to the addition of platelet activation inhibitors to solutions used in the pathogen reduction and subsequent storage of platelets. More particularly, the invention relates to the addition of adenylate cyclase stimulators and phosphodiesterase inhibitors to a platelet pathogen reduction and storage solution.
Reduction Of Contaminants In Blood And Blood Products Using Photosensitizers And Peak Wavelengths Of Light
Methods and apparatuses are provided for inactivation of pathogens in fluids containing blood products. Preferred methods include the steps of adding an effective, nontoxic amount of a photosensitizer such as riboflavin to the blood product and exposing the fluid to light having a peak wavelength.

This invention provides methods and compositions for using nitric oxide in a photoradiation pathogen inactivation process for whole blood and blood components to improve pathogen kill and to improve preservation of the quality of the blood components. This invention provides methods for using nitric oxide in combination with oxygen, photosensitizers, quencher and/or glycolysis inhibitor, and compositions comprising blood components decontaminated by these methods. Nitric oxide is provided using nitric oxide gas, or nitric oxide generators such as L-arginine, and/or N-acetyl-cysteine. This invention also provides compositions suitable for photoradiation pathogen inactivation that include fluid comprising a blood component, a photosensitizer, and dissolved nitric oxide. This invention provides decontamination systems useful for performing the methods of this invention and methods for making the decontamination systems. This invention also provides methods for decontaminating fluids and methods for increasing the storage life and quality of photochemically decontaminated platelets.
