Abstract:

The field of the present invention pertains to a controlled transfer biological collection device using a dry solid storage and transfer medium and a method for the collection of biological material of interest (genetic or proteinaceous material) in a form suitable for storage and/or subsequent analysis. Specifically, the present invention provides for a sampling device that controls the transfer of the biological sample to the storage medium by holding the storage medium and a moveable sample collection member having an analyte collection surface. The invention further provides for a method not only for storing a biological analyte on this collection device but also for analyzing the stored biological analyte using methods that are suited for automated analyzing systems.

Abstract:

The present invention relates to a novel device for separating plasma from a whole blood sample on a treated microporous membrane. An advantage of the present invention is that the sample can be assayed on the membrane after separation has occurred. A lateral flow chromatographic assay membrane has a sample application zone, a separation zone, and an analysis zone. An erythrocyte binding agent is added to the membrane in an amount sufficient to bind any erythrocytes present in the sample. Methods for using such devices are disclosed also.

Abstract:

The present invention relates to a device for preparing assay samples using a number of microscope slides. Each slide has a number of assay reaction surface locations spaced on the planar surface of the slide. In preferred embodiments, the device comprises, in part, a microscope slide holder that has the exterior dimensions of a SBS standard microplate, such as a 96 well plate. The device accepts conventional microscope slides equipped with sixteen microarray surfaces spaced nine millimeters apart on center, or four for a 96 well plate. Individual chamber plates are placed on top of the slides, creating an individual well above each assay reaction surface location. In preferred embodiments, each assay reaction surface location can comprise a microarray of multiple reactive sites. Thus, parallel processing can be done of samples for genomic or proteomic profiling. An advantage of the present invention is that one can use the conventional high throughput assaying equipment for SBS standard microplates while using conventional microscope slides, thereby allowing the use of robotic assay reading equipment designed for slides.

Abstract:

The present invention relates to protein microarray devices having an internal calibrator, kits containing such devices, and methods of using such devices. Such a device comprises a plurality of protein arraying pads on a support substrate, a plurality of protein arraying spots on each pad, and a calibrator disposed on each pad, each calibrator comprising a series of spots of increasing concentration of a protein. In preferred embodiments, each pad has an immunoglobulin calibrator that is of the same species as the proteins that are reactive to the protein arraying spots. An advantage of the present invention is that one can use reliably use microporous surfaces for conducting multiplexed protein microimmunoassays with an assurance as to surface-to-surface fluorescence variability on a microarray device.

Abstract:

The invention provides a method for supplying an enzyme substrate to a membrane-based, enzyme-linked reaction, comprising providing an open pore, high liquid retention capacity material impregnated with a predetermined amount of a substrate for the enzyme; and contacting the material with a membrane containing the enzyme-linked reaction under conditions which permit diffusion of the enzyme substrate to sites on the membrane containing the enzyme linked reaction. Abstract:

The present invention relates to devices and methods for the collection, storage, and purification of nucleic acids, such as DNA or RNA, from fluid samples for subsequent genetic characterization, primarily by conventional amplification methods. The present invention can be used to collect, store, or purify nucleic acids either from a biological source other than untreated whole blood, the biological source having naturally occurring nucleic acid amplification inhibitors present, (including either a buccal swab, cerebrospinal fluid, feces, lymphatic fluid, a plasma sample, a saliva sample, a serum sample, urine, or a suspension of cells or viruses), or from a treated whole blood source that has naturally occurring nucleic acid amplification inhibitors present, as well as added blood stabilization components that also inhibit nucleic acid amplification. More importantly, these nucleic acids can be released after collection or storage in a manner that enables them to be amplified by conventional techniques such as polymerase chain reaction. In particular, an absorbent material that does not bind nucleic acids irreversibly is impregnated with a chaotropic salt.
