Abstract:
The presence of clonal macrophages in pre-cancerous and cancerous tissue represents an early stage of the disease in which clonal expansion of macrophages occurs due to HIV integration or other genetic mutation. Clonally expanded macrophages induce proliferation of surrounding tissue leading to cancerous tumor growth. The invention provides methods and kits for diagnosis of HIV- and non-HIV-associated clonal expansion of macrophages in pre-cancerous and cancerous tissue. The invention also provides methods for the treatment of cancers induced by clonal macrophage expansion and proliferation of surrounding tissue.
Evaluating And Predicting Clinical Outcomes By Gene Expression Analysis
Methods of determining clinical outcomes in patients suffering from a pathological condition or syndrome are provided. Levels of intracellular gene expression re measured from a clinical sample provided by the patient, and the levels are compared to reference levels. Deviations from reference levels are predictive of clinical outcomes, for example, disease progression or response to therapeutic intervention.

Methods for modulating macrophage proliferation in an individual afflicted with or at risk for a macrophage-associated disease are provided. The methods employ a polyamine analog, or salt or protected derivative thereof. Macrophage proliferation has been implicated in a number of serious disorders, including AIDS (HIV)-associated dementia, AIDS-associated non-Hodgkin's lymphoma, and Alzheimer's disease. The invention also provides methods for aiding diagnosis and monitoring therapy of a macrophage-associated non-HIV associated dementia, especially Alzheimer's disease. The invention also provides methods of delaying development of macrophage-associated non-HIV associated dementias, including Alzheimer's disease, which entail administration of an agent which modulates macrophage proliferation.
Chlorite In The Treatment Of Neurodegenerative Disease
The invention features methods of treating a macrophage-associated neurodegenerative disease such as amyotrophic lateral sclerosis (ALS), Alzheimer's disease (AD), or multiple sclerosis (MS) in a subject by administering chlorite in an amount effective to decrease blood immune cell activation. A latch apparatus is configured to latch an actuator arm assembly that can position a read/write head relative to a data storage disk in a disk drive. The latch apparatus includes a latch arm, a coil, a permanent magnet, and a latch control circuit. The latch arm is configured to be moved between a closed position that inhibits movement of the actuator arm assembly in a landing zone and an open position that does not inhibit movement of the actuator arm assembly in the landing zone. The coil is configured to generate an electromagnetic force on the latch arm responsive to a coil current. The permanent magnet is configured to generate a magnetic force on the latch arm that is substantially opposed to the electromagnetic force from the coil. The latch control circuit is configured to increase according to defined open current profiles over a defined opening time period the coil current to at least an upper defined level to cause the coil to move the latch arm from the closed position to the open position. The latch control circuit is further configured to decrease according to defined close current profiles over a defined closing time period the coil current to no more than a lower defined level to cause the permanent magnet to move the latch arm from the open position to the closed position.
Methods For Treating Viral Infections Using Polyamine Analogs
Methods for treating viral infections using polyamine analogs, including mitoguazone (MGBG), are provided. In these methods, polyamine analogs destroy macrophages that act as viral reservoirs, facilitating the destruction of the viruses that dwell within the macrophages. Examples of viral infections that may be treated with the present methods include, but are not limited to, infections from human immunodeficiency viruses. These methods differ from previous methods of treatment using polyamine analogs, wherein the polyamine analogs were administered only as anti-tumor agents.
