Abstract:

Provided is a composition comprising a Vpr polypeptide conjugated to a therapeutic molecule. Preferably, the Vpr comprises synthetic Vpr. The therapeutic molecule can comprise any molecule capable of being conjugated to Vpr or a fragment thereof, including a polypeptide, a polynucleotide, and/or a toxin. The invention additionally provides a method for delivering a molecule into a cell. The method comprises contacting the cell with a conjugate comprising a Vpr polypeptide conjugated to the molecule. The invention further provides a method for modulating the expression of a transgene in a cell, a method for killing a target cell population in a subject, a method for increasing the sensitivity of cells to radiation therapy, and a method for inhibiting cell proliferation.

Abstract:

The present invention provides screening methods for identifying a compound that induces loss of the lentiviral protein Vpr; screening methods for identifying compounds that inhibit the peptidyl-prolyl cis/trans isomerase (PPIase) activity of a protein that catalyzes cis-trans isomerization of cis-peptidylprolyl bonds in Vpr; and compounds identified by the screening methods. The compounds are useful for treating a lentiviral infection. The present invention further provides methods of inducing loss of the lentiviral protein Vpr; methods of inhibiting lentivirus viral replication; and methods of treating a lentivirus infection in an individual. The methods generally involve administering to an individual infected with the lentivirus an effective amount of a compound that induces Vpr loss and/or that inhibits PPIase activity of a protein that catalyzes cis-trans isomerization of cis-peptidylprolyl bonds in Vpr.

Abstract:

Variant forms of chimeric protein molecules comprising a Factor VII moiety and an Fe region of an IgG1 moiety provide improved properties. The variants are more resistant to proteolytic degradation. Thus preparations of the variant forms are more homogeneous and have a longer half-life. The variant forms are used for treating cancer, atherosclerosis, psoriasis, diabetic retinopathy, wet macular degeneration, and rheumatoid arthritis.

Abstract:

A new spinal hook of the present invention which comprises a hook-configured shoe with an integral portion. The top portion includes a pair of central posts which are displaced to form a slot therebetween. Each central post includes a groove colinearly formed to receive a spinal rod within. The slot between the two posts is oriented with respect to the grooves so that an eyebolt assembly may be used to engage the hook to the fixation rod. In another aspect of the invention, a slot is formed in the end faces of the hook. The slot forms one part of a tongue and slot instrumentation engagement arrangement, with the hook-holding instrument including a correspondingly configured tongue. In another aspect of the invention, each of the pair of central posts include grooves on both lateral faces of the posts so that the spinal rod can be situated on either side of the spinal hook.

Abstract:

A lateral offset connector is provided for engaging a spinal rod to a vertebral fixation element, such as a hook or a bone screw. The lateral offset connector includes a head portion from which a pair of parallel arms extend to form a slot opening between the arms. The arms are configured with grooves on their underside for receiving the spinal rod. The slot opening between the arms is configured to receive an eyebolt engagement assembly therebetween. The eyebolt is disposed between the arms of the connector and a nut is threaded onto the threaded post of the eyebolt to clamp the spinal rod between the eyebolt and the offset connector arms. The offset connector further includes a threaded post extending from the end of the connector for engagement with the spinal fixation component, such as the bone screw or hook. A nut clamps the fixation component between a rounded shoulder of the connector and the nut. The plurality of grooves in the arms of the connector provides means for readily engaging a plurality of vertebral fixation elements to a spinal rod where the the fixation elements are not colinear along the vertebral column, that is where the lateral distances of the fixation elements relative to the spinal rod varies along the length of the rod.
