Abstract:

The present invention relates to drug screening assays and methods for the treatment of proliferative disorders associated with elevated levels of heat shock protein 72 (Hsp72) expression in a cell. The invention is based on the discovery that overexpression of full length Hsp72 protein, or the C-terminal protein binding domain of Hsp72, results in oncogenic transformation of cells.

Abstract:

A device for mixing, homogenization, resuspension, disintegration, disruption, or centrifugation of media placed in test tubes, including: a rotor having a replaceable closure; a driver having a substantially vertical shaft; a sleeve having a spiral groove and a bush moveable along the sleeve, moving the rotor up and down; and a stationary ring with extended teeth faced toward the rotor. When the rotor is in its lower position the tubes engage the teeth and receive strikes which induce vigorous motion and effective mixing of media inside the tube. When the rotor is in its upper position the tubes are disengaged with the teeth and rotation of the rotor provides centrifugation of the media.

Abstract:

The present invention is based on the discovery that inactivation of the heat shock response in cancer cells significantly enhances their sensitivity to proteasome and Hsp90 inhibitors. The inventors have discovered novel compounds which exhibit low toxicity, inhibit the heat shock protein response and sensitize cancer cells to anti-cancer therapies. In general, the heat shock protein inhibitors of the present invention share a common structure, namely a 2H-benzo[a]quinolizine tricyclic ring. Also encompassed are methods for a high throughput screen to identify heat shock inhibitors that sensitize cancer cells to anti-cancer therapies.

Abstract:

Disclosed herein is a method for inducing Hsp72 production in a cell. The method involves contacting the cell transiently with a proteasome inhibitor in an amount sufficient to induce Hsp72 production. Preferably Hsp72 levels are induced to a level sufficient for the suppression of stress-activated kinase activity attributable, for example, to a stress-activated kinase selected from the group consisting of JNK and p38. The method has both in vitro and in vivo applications. Also disclosed is a method of the type described above is applied specifically to aged cells. Also disclosed is a method for treating pathologies associated with apoptosis and inflammation in an individual. The method involves administering to the individual a proteasome inhibitor in an amount sufficient to induce Hsp72 production. In a preferred embodiment of the therapeutic method, the individual is an aged individual. Stress-induced pathologies such as ischemia are particularly important with respect to the therapeutic aspect of the invention.

An auxiliary holder of tubes intended for disintegration of living cells, such as yeast cells, in the presence of glass beads, as well as for mixing substances and pellet resuspension. The holder is installed on a vibrator. Due to specially-made gaps between the tubes and the main parts of the holder the processed cells and substances are subjected to a complex impact of vibration created by the vibrator and shocks created by the holder. A wireless, dynamically reconfigurable link layered communications network and method are disclosed. A destination transceiver such as, a central transceiver, has a unique, permanent identification with a predetermined numerical link layer level n. A plurality of remote transceivers each having a unique, permanent identification and a variable, stored numerical link layer x where x>n and x and n are integers and x-n-1 represents the number of remote transceivers through which a communication must pass to reach the destination transceiver. The variable, stored numerical link layer level of each remote transceiver is dependent upon the dynamic condition of the network at the time of message transmission from or through the particular remote transceiver.
