    A method and apparatus for controlling a stream of liquid and air segments wherein the liquid and air segments are selectively aspirated into a first fluid conduit in a plurality of cycles, each cycle beginning with the aspiration of a first air segment and ending with the aspiration of a final air segment. The liquid and air segments are then transferred from the first fluid conduit to a second fluid conduit. The volume of the final air segment of each cycle is then adjusted after the final air segment has moved into the second fluid conduit. Next, the liquid segments and the air segments of each cycle are transferred from the second fluid conduit to a third fluid conduit. The volume of the first air segment of each cycle is then adjusted after the first air segment has moved into the third fluid conduit.

  • Abstract:
    An apparatus and a method for analyzing a plurality of samples according to an analysis protocol is described, wherein each of the samples is contained within a liquid test package comprising a plurality of liquid and air segments including a reagent segment and wherein during at least a portion of the protocol the reagent segment emits photons. At least one of the liquid test pack includes a buffer segment separated from the reagent segment by a vanish bubble. The apparatus comprises a fluid conduit through which the liquid test packages are flowed, a first luminometer positioned along the fluid conduit for detecting photons emitted by the reagent segment of the at least one of the liquid test packages, a second luminometer positioned along the fluid conduit for detecting photons emitted by the reagent segments of the liquid test packages other than the at least one of the liquid test packages, and a vanish zone contained within the fluid conduit between the first and second luminometers. The reagent segment and the buffer segment of the at least one of the liquid test packages are mixed together in the vanish zone such that the light/photon producing reaction is substantially ceased. The method comprises the steps of inserting the liquid test packages into a fluid conduit, flowing the liquid test packages past a first luminometer positioned along the fluid conduit and detecting photons emitted by the reagent segment of the at least one of the liquid test packages, mixing the reagent segment and the buffer segment of the at least one of the liquid test packages after the photons have been detected, and flowing the liquid test packages past a second luminometer positioned along the fluid conduit and detecting photons emitted by the reagent segments of the liquid test packages other than the at least one of the liquid test packages.

    A random access, high-throughput system and method for preparing a biological sample for polymerase chain reaction (PCR) testing are disclosed. The system includes a nucleic acid isolation/purification apparatus and a PCR apparatus. The nucleic acid isolation/purification apparatus magnetically captures nucleic acid (NA) solids from the biological sample and then suspends the NA in elution buffer solution. The PCR testing apparatus provides multiple cycles of the denaturing, annealing, and elongating thermal cycles. More particularly, the PCR testing apparatus includes a multi-vessel thermal cycler array that has a plurality of single-vessel thermal cyclers that is each individually-thermally-controllable so that adjacent single-vessel thermal cyclers can be heated or cooled to different temperatures corresponding to the different thermal cycles of the respective PCR testing process.

    A method and reagent composition provide for the removal or blocking of serum interferences in liposome immunoassays. A carrier or liposome exhibiting at least one domain of phosphoryl ester groupings different from those of the marker liposome is introduced into the reaction mixture, whereby endogenous interfering species competitively bind to such carrier or liposome rather than to the marker liposome. Also, soluble phosphoryl ester groupings can be introduced into the reaction mixture which react with such endogenous species and further reduce serum interferences.

    Improved assay methods utilizing stable, enzymatically triggered chemiluminescent 1,2-dioxetanes with improved water solubility are provided. Assays are performed by a capsule chemistry analytical assay method wherein fluid capsules comprising discrete aqueous segments containing either a chemiluminescent dioxetane reagent or an activating agent separated by an oil-based isolating fluid are flowed through a conduit, and subsequently reacted to produce light. The improvement comprises using a dioxetane substituted with two or more water-solubilizing groups disposed on the dioxetane structure to provide the chemiluminescence. Compositions comprising such a dioxetane, a non-polymeric surfactant enhancer and optionally a fluorescer, provide enhanced chemiluminescence and eliminate the problem of reagent carryover when used in assays performed on capsule chemistry analytical systems.
