Abstract:
Triglycerides in biological fluids containing interfering substances, such as phospholipids, are determined by hydrolyzing the triglycerides to glycerol in a short time span at moderate temperatures with a solution of alkali metal hydroxide in methanol, and treating the resultant hydrolysis mixture with a water soluble magnesium salt to precipitate the interfering substances. Glycerol is then colorimetrically determined in the resultant supernatant solution by incubating with a single reagent-substrate composition to develop a measurable color.

A novel reagent and method for colorimetrically determining the concentration of triglycerides in biological fluids, and for rapid determination of glycerol are described. Triglycerides are hydrolyzed in methanolic base to glycerol, and glycerol content is determined colorimetrically.
