Abstract:

An efficient transdermal delivery system for delivering an active ingredient to the blood supply of a living body, comprising a vasodilator, an active ingredient, and a permeation enhancer for the active ingredient. The components of this mixture are combined in a manner that dissolves them into solution, eliminating the need for a polymeric binding agent and thus diminishing the total molecular weight of the mixture and increasing the penetration and delivery efficiency of the active drug molecules.

A method for treating a patient suffering from a wound, ulcer, or inflammation includes providing a topical formulation comprising a carrier and a combination of active ingredients that includes at least one retinoid and at least one blood vessel dilator, and applying the topical formulation to the skin of the patient to cause healing, accelerated healing or prevention of the wound, ulcer or inflammation. Abstract:

The synthesis and use of bifunctional molecules to improve the topical and transdermal delivery efficiency of various types of therapeutic agents or agents designed to promote the transdermal delivery of those therapeutic agents either into the skin tissue or into the systemic circulation. Three major classes of molecules are covalently joined as bifunctional substances; chemical vasodilators, passive dermal penetration enhancers and therapeutic or diagnostic drugs. Chemical vasodilators may be delivered into the skin to increasing the blood flow in a tissue that has compromised circulation or they may be used as part of a delivery vehicle to promote the delivery of the drug. Passive dermal penetration enhancers are those chemicals that promote the passive penetration of drugs and other chemicals through the stratum corneum and epidermis of the skin tissue. Drugs and diagnostic agents are the third group of chemicals that are candidates for the linkage of molecules.

Abstract:

A system for the delivery of chemical vasodilators into the skin for the purpose of increasing the blood flow in a tissue that exhibits compromised circulation. The designated purpose of this increased blood flow is to transiently reverse or inhibit or prevent the onset of an associated medical affliction that may in part or in whole, be either the consequence of that medical condition or participating in the clinical symptoms and etiology of that medical condition. The process and the effect of the topical delivery of a chemical vasodilator has been previously demonstrated for other medical and pharmaceutical applications, however the specific use of this class of drug molecules for the focused effect of improving blood flow to the skin and surrounding tissue with the goal of improving the metabolic health of the tissue and to possibly affect the symptoms of a medical condition is the specific aim of this patent.

Abstract:

A method for determining and demonstrating the role of vasodilator chemical agents in the development and practice of transdermal drug delivery systems. Vasodilator chemicals applied topically dilate the blood vessels in the skin tissue, which have been shown to facilitate or inhibit systemic or skin tissue deposition of drug substances. The level of stimulation and/or inhibition has been found to be dependent on the concentration and the identity of the specific vasodilator chemical(s) used as well as the drug molecule(s) to be delivered. This work teaches the need to consider specific formulation requirements when dealing with vasodilator chemicals for the creation of successful delivery vehicles in the transdermal drug delivery system. These requirements for very low concentrations of vasodilators were an unexpected and a surprise finding, in contrast to the concentrations of the vasodilators typically used to elicit an increase in skin blood flow.

Abstract:

A formulation and method for the preparation of topical anesthetics formulations, containing one or more anesthetics, in a delivery vehicle designed to promote penetration of the anesthetics into the skin and underlying tissue and to facilitate the speed of action and prolonged response. The stratum corneum layer of the skin at the application site is at least partially removed by an abrading substance, such as pumice, preferably incorporated into the formulation, or by a step that uses a mechanical abrasion of the outer layer of skin, such as a rotating or vibrating brush. The enhanced penetration into the live skin tissue is achieved by removing the outer layer of stratum corneum skin tissue by including an abrading substance in a formulation containing the anesthetics, then rubbing the formulation into the skin, or the use of a brush-like device to assist in the removal of the skin layer.
