A method for detecting and/or quantifying a target DNA or RNA present in a sample by a isometric primer extension method is disclosed. The method includes carrying out a primer extension reaction in the absence of a free nucleotide so that the primer extension reaction is stopped where the absent nucleotide would have been inserted. Thus, as the amount of incorporation of a labeled nucleotide on the primer extended product is detected, the amount of the target RNA or DNA in the sample is measured.

Abstract:
The present invention provides compositions and methods for inhibiting, reducing, preventing or controlling dental caries formation. Electrolyzed water having an oxidation-reduction potential from about -150 to 0 mV measured against a platinum electrode, also known as “Trim water” can reduce, inhibit, prevent, or control caries in part by neutralizing dental plaque acid by increasing the pH in the area surrounding dental plaque. One embodiment provides contacting a tooth with electrolyzed water, typically after the tooth has been challenged with a sugar, to neutralize dental plaque acid and thereby reduce the formation of dental caries. Electrolyzed water and compositions comprising electrolyzed water can be formulated as palatable beverages, gels, candy, or other foodstuffs that are safely digested by individuals such as young children.
Isometric Primer Extension Method And Kit For Detection And Quantification Of Polynucleotides
Methods, compositions, and kits for the detecting and/or quantification of polynucleotides are provided. An exemplary method provides hybridizing a first primer to the target polynucleotide and forming equal length primer extension products using a nucleotide reaction mixture consisting of X, Y, and Z, wherein X and Y are different purine non-terminator nucleotides, and Z is a pyrimidine non-terminator nucleotide, or X and Y are different pyrmidine non-terminator nucleotides, and Z is a purine non-terminator nucleotide. A portion of the extension products are hybridized to a second primer that is immobilized on a solid support, and the second primer is extended with at least one nucleotide having a detectable marker. The signal from the detectable marker can be correlated to the amount of target polynucleotide. Kits for performing this method are also provided.
Detection Of Nucleic Acid Variation By Cleavage-Amplification (Cleavamp) Method
The invention relates to a method for detecting any mutation at a predetermined site occurring in a known nucleic acid sequence. The method uses primer extension analysis to detect the mutation. Unlabeled terminator is supplied along with labeled non-terminator in the primer extension reaction to detect whether the first nucleic acid base on the template strand that is directly opposite the nucleic acid base immediately 3′ to a primer is a mutant. In the primer extension reaction, the terminator is complementary to the wild-type base on the template strand that is directly opposite the nucleic acid base immediately 3′ to the primer. Non-terminators are the other nucleotides and are labeled. When the terminator is incorporated into the primer extension strand, primer extension reaction terminates. Incorporation of a labeled non-terminator in the primer extension strand indicates that a mutation has occurred at the predetermined nucleic acid base site.
Method For Identifying Gene Fusions By Circle Cdna Amplification

International Classification:
C12N 15/10 C12Q 1/6853
Abstract:
One embodiment provides methods to identify known and unknown gene fusions by creating a cDNA circle and analyzing the circle cDNA by amplification or sequencing. The circle cDNA is created in two approaches: 1) reverse transcribe a target RNA to cDNA, ligate the 3′end of the cDNA to its 5′ end to form a circle cDNA, or 2). ligate the 3′ end of target RNA to its 5′ end to form a circle RNA, reverse transcribe the RNA to a cDNA, and ligate the cDNA to form a circle cDNA. The circle cDNA is amplified using a primer designed from a known sequence of a wild type target gene by rolling circle amplification or PCR methods. The known or unknown fusion gene sequences in the circle cDNA are amplified and identified by sequencing analysis.

Abstract:
Systems and methods for MBIR reconstruction utilizing a super-voxel approach are provided. A super-voxel algorithm is an optimization algorithm that, as with ICD, produces rapid and geometrically agnostic convergence to the MBIR reconstruction by processing super-voxels which comprise a plurality of voxels whose corresponding memory entries substantially overlap. The voxels in the super-voxel may also be localized or adjacent to one another in the image. In addition, the super-voxel algorithm straightens the memory in the “sinogram” that contains the measured CT data so that both data and intermediate results of the computation can be efficiently accessed from high-speed memory and cache on a computer, GPU, or other high-performance computing hardware. Therefore, each iteration of the super-voxel algorithm runs much faster by more efficiently using the computing hardware.
