Abstract:

Described herein are a synthetic cascading bistable switches (Syn-CBS) circuit. In some aspects, the SynCBS circuit is single-strain circuit with two coupled self-activation modules to achieve two successive cell fate transitions. In other aspects, the SynCBS circuit is a two-strain circuit where the self-activation modules are divided in two cells instead of being expressed in a single cell. Also described are plasmids encoding the Syn-CBS circuits.

Abstract:

Provided herein are compositions and methods for real-time identification and isolation of base-edited cell populations. Also provided herein are methods for producing enriched isogenic lines of genetically modified cells, including base-edited human pluripotent stem cells. In particular, provided herein are methods utilizing transient expression of reporter proteins, the detectable signal of which is altered following base editing. Using the transient reporter with a base editor permits enrichment of isogenic populations of base-edited cells.

Provided herein are tools for understanding and engineering dynamics of synthetic genetic circuits which utilize CRISPR components. More particularly, methods, systems, and compositions for directing Cas9 activity using a fluorescent guide RNA (fgR-NA) which fluoresces in the presence of small molecules (e.g., DFHBI-IT) are described and illustrated in the present provisional application.
